Mark Sequences as passed
Set the read direction for all sequences by using the same procedure as used for setting the read direction of the trace files: Sequences → Set Read Direction. Select an arbitrary direction and click OK.
After that, mark all sequences and click on the Biocode button and then on Annotate with FIMS/LIMS Data.
Enter the correct plate name and click OK → the sequences in Geneious will now be updated with all changes which were made in the meantime in the DC (new identification will be updated in the Taxon_Name column and all flags (Checks, Contaminations etc.) will appear in the Problems column.
Transfer Barcodes back to Biocode LIMS
As next - and more or less final- step we pass the Barcodes and all the corresponding lab information back into the DC. For this, mark all consensus sequences which are NOT flagged as “CON/XCON/PCON” and click on the Biocode icon and then on Mark as Pass in LIMS. In the pop-up window insert your name and the name of the plate.
If you now go back into the corresponding sequencing plate, the wells of the passed sequences are now marked green and the barcode is attached to the well (double click on well, click View 1 sequencing results).
The next day the barcode and all the lab information which were entered on the plates are also attached to the corresponding DC entry.
