====== Download the Raw Sequences into Geneious ====== A **Geneious Shared Database** is set up for the CaBOL project. Please refer to the [[geneious:db_login|Geneious for CaBOL - Installation and Database Login]] guide to learn how to connect to the shared database. ==== Geneious Shared DB Folder Structure: ==== After connection you should create, establish, and stick some kind of recognizable folder-structure. We do it plate-wise like this: {{:geneious:image21.png|}} Create the folder for the plate in the format ''CaBOL_xxxx (organism group)'' and a sub folder ''Raw data''. Select this sub folder. Click on the **Biocode** icon in the navigation bar of Geneious and then on **Download Traces from LIMS**. {{:geneious:image22.jpeg|}} {{:geneious:image23.png|}} In the pop-up window, **enter the names of the two sequencing plates**. The raw sequences are now loaded into your raw traces folder and appear in the main window. {{:geneious:image24.jpeg|}} In the Geneious navigation bar, select **Tools**. Under **Sequencing**, select the appropriate binning profiles for vertebrates or invertebrates (Nextcloud folder **Geneious**; [[https://oc.zfmk.de/index.php/f/831677|https://oc.zfmk.de/index.php/f/831677]]; files: ''GBOL_invert.binProfile'' and ''CaBOL_vert.binProfile''). {{:geneious:image25.png|}} {{:geneious:image26.png|}}{{:geneious:image27.png|}}
